Symplex™ is Symphogen’s proprietary process for discovering polyclonal antibodies with diversity and specificity customized to a particular therapeutic application. The Symplex™ discovery process begins by isolating B lymphocytes, which are responsible for making antibodies, from naturally immune human donors displaying a high number of antibodies against a particular target antigen. The antibodies are then obtained from this starting material using plasma cell isolation and Symplex™ PCR cloning.

A critical problem solved by the Symplex™ technology is the direct isolation of antibody encoding genes from human immune starting material such that the original pairing of the antibody-building blocks, the heavy chains and the light chains, is completely maintained. Therefore, the high antibody affinities generated in the natural human immune response translate directly and unaltered into lead antibody compositions discovered by Symplex™.

Antibody leads are identified from compositions of original antibody gene pairs (known as cognate pairs) by high throughput screening for antigen specificity.

The Symplex™ technology provides potent and safe drug leads, because the antibodies have been generated in humans and mimic the natural immune response. All processes leading to the isolation of specific antibodies have been automated, thus, the Symplex™ technology is a fast method to generate naturally high affinity drug leads.

Another benefit of this technology is its ability to identify not only polyclonal, but also monoclonal antibody drug leads, depending on the complexity of the target.
